P1NP ELISA Kit (Procollagen Type 1 N-Terminal Propeptide)
INTENDED USE
Human P1NP ELISA kit can be used for measuring levels of procollagen type 1 N-terminal propeptide (P1NP) using human serum and plasma.
INTRODUCTION
Procollagen type 1 N-terminal propeptide (P1NP) has two distinct forms in humans. Its presence aids in the diagnosis of several conditions including osteogenesis imperfecta and Ehlers-Danlos syndrome that are brought on by a lack of collagen. The gene contains six exons, spanning over more than 19 kb of DNA. The protein encoded by this gene is composed of four domains: a N-terminal propeptide domain, a collagen domain, a proline/hydroxyproline rich region and an end peptide domain. The collagen domain is of particular importance as it serves as the major binding site for type I procollagen molecules during intracellular assembly. P1NP is a membrane-anchoring protein that promotes cell adhesion and regulates fibronectin production. P1NP expression varies widely among cancers, making it difficult to use it as an easy marker for cancerous cells.
P1NP contains three different domains: (1). the 5-kDa proline-rich region N-terminal domain (D5KRP), which has been found to be important for collagen synthesis and formation of collagen fibril structures; (2). the proline-rich region central domain (D4CR1), which is responsible for binding to collagen and (3). the helix-loop-helix domain (D3HHL), which is responsible for binding collagen fibrils.
P1NP is a biomarker of bone formation and elevated levels can indicate increased bone turnover, this can lead to osteoporosis and fractures. It is important to monitor levels to prevent these conditions from becoming more severe. It is an acute phase reactant. This means that it responds to inflammation and other responses from the immune system.

All reagents supplied need to be stored at 2 °C – 8 °C, unopened reagents will retain reactivity until expiration date. Do not use reagents beyond this date.

TYPICAL RESULTS
For the following P1NP ELISA kit it is recommended that a standard curve is generated for each assay carried out.
Standard Curve: 0, 62.5, 125, 250, 500, 1000, 2000, 4000 pg/ml.
