INTENDED USE
Human TGF-beta ELISA kit can be used to detect concentrations of transforming growth factor beta (TGF-beta, TGF-β1, CED, DPD1) present in human serum, biological fluids, tissue homogenates and plasma samples. This assay has a minimum sensitivity detection limit of 18.75 pg/ml.
INTRODUCTION
Transforming growth factor beta (TGF-beta) is involved in the regulation of cell proliferation and differentiation, it also plays an important role in wound healing and tissue repair processes. It has been shown to be a possible therapeutic target in the treatment of metabolic syndrome, Type 2 diabetes mellitus and vascular complications. The protein is initially synthesized as a precursor molecule that is called pro-TGF-beta, it is then cleaved into the active forms of TGF-beta1, TGF-beta2 and TGF-beta3. It is mainly produced within the intestines, whilst both TGF-beta2 and TGF-beta3 are produced somewhere elsewhere.
The effects of TGF-beta1 mainly occurs in the gut, liver, lungs and kidneys. Whist those of TGF-beta2 and TGF-beta3 it only occurs in the liver and kidneys. It is secreted by a variety of cells, including fibroblasts, keratinocytes and inducible T cells. In addition to its mitogenic effects on progenitor cells and other mesenchymal tissues, it is also an important factor in the development of bone tissue (osteoblasts). It plays a key role in senescence-associated osteoclasts resorption of bone matrix by stimulating p38 MAP kinase signalling pathway which leads to the release of calcium from the mineral compartment, a necessary step for bone resorption.
In the eye, TGF-beta it is secreted by photoreceptors and choroid to maintain the basement membrane and glaucoma filaments, respectively. In the central nervous system, it prevents granule cell death during olfactory bulb development in mice. Alternatively, it has been shown to preserve axotomized hippocampal neurons in culture via CCN3 gene expression.
