BACKGROUND

IGFBP2 (insulin like growth factor binding protein 2) is a 31.3 kDa polypetide protein that is unglycosylated and does not circulate with a circadian rhythm. It also forms binary IGF-complexes. While the concentration in serum increases during fasting, after trauma, after major surgery and in malignant conditions, the concentration in blood is age-dependent and its concentration decreases with age. It belongs to type I membrane-bound growth factor binding proteins and interacts with tissue growth factors as well as regulates their release from cells, which affects physiological functions such as proliferation, differentiation and apoptotic cell death. IGFBP2 (IGFBP2a and IGFBP2b) has two isoforms in humans. The shorter isoform is the major protein isoform, but the longer form is seen during development in fetuses and infants. These are differentially expressed in cancer patients. This difference of expression may be due to differences in metabolism or function.

It is especially intense in malignant conditions, where the normalisation of serum levels during remission and the association of the IGFBP-2 level to the degree of progression can be startling features in a variety of tumour types. In cases of GH usage (doping) and short stature, for instance, it has been discovered that levels fall during GH-therapy. Moreover, maternal serum levels are lower and IGFBP-1 levels are higher in trisomy 18. As a result the IGFBP-2/IGFBP-1 ratio can serve as a reliable signal or marker for chromosome abnormalities. The expression of the receptor protein itself is also regulated by IGFBPs. In areas of high IGFBP levels, the expression and signalling of the receptor is kept in check. The binding of both soluble and insoluble forms to cell membrane can exert a negative feedback on the receptor’s function.

INTENDED USE

Rat/Mouse IGFBP-2 ELISA kit is designed for analysing quantitative concentrations of mouse or rat insulin-like growth factor binding protein-2 (IGFBP2) using samples of serum. This assay has a minimum sensitivity detection limit of 0.01 ng/ml.

The standard range of insulin-like growth factor binding protein-2 (IGFBP2) using current rat/mouse IGFBP-2 ELISA kit was approximately 0.01 ng/ml. The dynamic range for this assay is 31.25 – 1,000 pg/ml.
