INTENDED USE

Human Protein Wnt-7a ELISA kit can measure concentrations of WNT7A (Protein Wnt-7a) present is plasma, serum, biological fluids or tissue homogenate samples.

INTRODUCTION

WNT7A encodes a secreted lipid-modified signaling protein that activates the canonical Wnt/β-catenin pathway to regulate developmental patterning and tissue homeostasis. By stabilizing cytoplasmic β-catenin allowing its nuclear translocation, WNT7A elicits distinct transcriptional programs governing cell fate, motility, and polarity with impacts spanning embryogenesis, neurogenesis, and tumorigenesis when dysregulated. Intriguingly, WNT7A exhibits remarkably cell type-specific tumor suppressive or oncogenic effects contingent on cellular context.

In the developing female reproductive tract, WNT7A signaling specifies establishment of the Mullerian duct axis essential for reproductive tract integrity. Its absence severely compromises oviduct, uterus, cervix and upper vagina formation underlying subsequent infertility. Meanwhile in the central nervous system, WNT7A stimulation enhances dendritic branching, neuroplasticity and synapse formation critical for adaptive responsiveness, hippocampal-dependent memory and stress resilience.

However, epithelial WNT7A signaling elicits divergent consequences in cancers contingent on tumor lineage. In lung cancer, WNT7A suppression fosters tumor-promoting inflammation. Its restoration attenuates NF-kB-mediated cytokine release through novel cross-talk between DIg/β-catenin and IKKβ/RelA complexes. Contrastingly in certain pancreatic and ovarian tumors, upregulation advances pathogenesis by fueling proliferation. These starkly opposing outcomes reflect intricacies of intersecting signaling networks downstream of homeostatic WNT7A across distinct cell types.

For this WNT7A ELISA kit, it is recommended that a standard curve is generated for each assay carried out.
