INTENDED USE
Human NLRP3 ELISA kit can measure concentrations of nod like receptor pyrins-3 (NLRP3) present is plasma, serum, biological fluids or tissue homogenate samples.
INTRODUCTION
NLRP3, also known as NOD receptor pyrin domain containing 3 is a protein complex that plays a role, in the body’s immune responses. It belongs to a group of receptors called NOD receptors, which are found inside cells and can recognize signals associated with pathogens and potential harm.
Specifically, NLRP3 is a part of the inflammasome, which is a complex made up of multiple proteins that helps activate pro inflammatory cytokines like interleukin 1beta. When various signals such as toxins ATP (adenosine triphosphate) or crystalline substances trigger it comes together with proteins to form the inflammasome complex. This triggers caspase 1 activation leading to the conversion of pro IL 1beta into its active form. The release of IL 1beta then starts a response to fight against pathogens or tissue damage. However excessive activation of NLRP3 can cause inflammation. Has been associated with different autoimmune and inflammatory diseases.
While NLRP3 is primarily found in cells like macrophages, neutrophils, and dendritic cells it can also be present in other cell types such as epithelial cells, osteoblasts (bone forming cells) and neurons. This widespread distribution allows NLRP3 to detect danger signals indicating invasion or cellular damage, throughout the body.
Besides pathogens and molecules disruptions, in the release of potassium, calcium signaling, malfunctioning mitochondria and oxidative stress have the potential to activate NLRP3. This highlights the role, as a detector of responding to various internal and external threats.

For this NLRP3 ELISA kit, it is recommended that a standard curve is generated for each assay carried out.
