INTENDED USE

Human Humanin ELISA kit can measure concentrations of MT-RNR2 (humanin) present is plasma, serum, biological fluids or tissue homogenate samples.This assay has a minimum sensitivity detection limit of 0.12 pg/ml.

INTRODUCTION

First identified in 2001, humanin is encoded in the 16S rRNA gene within the mitochondrial genome. At only 24 amino acids in length, it is the smallest known peptide encoded by the human mitochondria. Despite its small size, it has significant cytoprotective effects that researchers are only beginning to understand.

The most striking evidence relates to Alzheimer’s disease – levels in the brain decline with the progression of Alzheimer’s, and administering exogenous humanin appears to rescue neurons and improve memory in animal models. This neuroprotective effect arises from it’s ability to interact with the amyloid precursor protein, thereby reducing amyloid beta toxicity. Humanin may confer similar protection in diabetes by interacting with the insulin receptor to trigger downstream signaling cascades.

Excitingly, pharmaceutical companies have already developed synthetic-humanin analogs that show enhanced potency and bioavailability compared to the natural peptide. Early clinical trials demonstrate these humanin-analogs are safe, well-tolerated, and achieve much higher circulating levels then native when administered. Larger scale phase 2 and 3 efficacy trials will follow to determine if humanin-therapy can prevent neuronal deterioration in early Alzheimer’s disease or help regulate glucose metabolism in diabetes. If successful, analogs may soon provide clinicians powerful new tools to combat these age-related diseases.
