INTENDED USE

Human caspase 1 ELISA kit can measure concentrations of CASP1 (caspase-1, ICE, CASP-1, ICEP45) present is plasma, serum, biological fluids or tissue homogenate samples. This assay has a minimum sensitivity detection limit of 46.875 pg/ml.

INTRODUCTION

Caspase-1 sits at the crux of inflammatory processes as the essential enzymatic executor enabling cytokine maturation and pyroptotic cell death – two cardinal responses against pathogens. First described in 1996 as the founder of inflammatory caspases, it took years of ensuing research to reveal mechanistic details underlying its influential role marshaling anti-microbial immunity. CASP1 becomes activated upon assembly of cytosolic multiprotein complexes termed inflammasomes, which recognize various pathogenic signatures through sensor proteins like NLRP3 or NLRC4.

Sensor oligomerization into an inflammasome platform enables recruitment and autocatalytic cleavage of CASP1 into active fragments. Activated caspase-1 then cleaves downstream substrates to drive inflammation on two fronts. First, it processes pro-interleukin (IL) cytokines like pro-IL1β and pro-IL18 into mature, secreted forms essential for lymphocyte activation and fever induction. Secondly, caspase-1 triggers pore-formation by gasdermin D to permeabilize the plasma membrane allowing IL1 cytokine release. Yet gasdermin pores also precipitate osmotic cell lysis, termed pyroptosis, eliminating intracellular niche for bacterial replication while leak of danger signals recruits more immune cells.

Thus pharmaceutical targeting of caspase-1 or upstream inflammasomes continues gaining attention. Cambinol, a recent inhibitor blocking NLRP3 inflammasome assembly, shows effectiveness reducing IL1β production in rodent models of arthritis and multiple sclerosis. Looking forward, strategies more selectively dampening CASP1 over other inflammatory caspases could minimize infection susceptibility.
