INTENDED USE

Human glucagon ELISA kit can detect amounts of glucagon (glicentin, GC) present is human plasma, serum, biological fluids and tissue homogenates. This assay has a sensitivity detection limit of 4.688 pg/ml.

INTRODUCTION

Glucagon is an important peptide hormone which is produced by the alpha cells that are found in the pancreatic islets, its main function is that of a counter-regulatory partner to the hormone insulin. While insulin functions in the lowering of blood sugar after eating a meal, glucagon is the antagonist and it is responsible for preventing the blood glucose levels from dropping too low, this is known as condition called hypoglycemia. However, when a body is in that state fasting state or when it is under intense physical stress, this causes the pancreas release glucagon into the bloodstream. The released glucagon mainly travels to the liver, where it is responsible for triggering glycogenolysis. This is the breakdown of stored glycogen into glucose and gluconeogenesis which is the synthesis of glucose from other non-carbohydrate sources such as amino acids. By the process of stimulating this pathway, glucagon is important is ensuring that the primary organs such as the brain and the muscles have a continuous supply of fuel (glucose), thereby maintaining the systemic homeostasis during periods when energy is in a deficit state.

Furthermore, to its important role in glucose mobilisation, glucagon has a significant influence on lipid metabolism and energy expenditure. Through the process of signalling the liver to shift away from fatty acid synthesis and to move towards fatty acid oxidation, glucagon can promote the production of ketone bodies. This is vital and it serves as another source of energy during period of prolonged fasting or when carbohydrate is restricted. This kind of metabolic flexibility is important for survival especially during periods of nutrient scarcity. Recently, glucagon has be known to have a role as a thermogenic and this can reduce appetite, many leading researchers are investigating its potential in therapies for weight management. In addition, when it is used in combination with the GLP-1 receptor agonists, glucagon-based treatments have been used with the aim to balance blood sugar control by increasing the caloric burn, this offers a more sophisticated approach in the treatment of obesity and various metabolic dysfunction.
