Sestrin-2 ELISA Kit (SESN2)

INTENDED USE

Human sestrin-2 ELISA kit can detect amounts of SESN2 (sestrin-2, Hi95, SEST2) present is plasma, serum, biological fluids and tissue homogenate samples. This assay has a minimum sensitivity detection limit of 0.094 ng/ml.

INTRODUCTION

Sestrin-2 belongs to the family of globin-like α-helix-fold proteins, consisting of 23 helices and no β-sheets. It functions in the regulation of cell growth and survival. It has a protective effect on physiological and pathological states mainly via regulating, oxidative stress, endoplasmic reticulum stress, autophagy, metabolism and inflammation. It displays 33% sequence identity to “globin-like” proteins. It is encoded by the SESN2 gene, which is a nonsense mutation causing the loss of all the original amino acids present in its translated form (the deletion removes all stop codons and three of four start codons). This deletion likely occurred during evolutionary history and has resulted in a protein without any functional regions. Humans and rats both contain this protein, but birds do not. Additionally, the SESN2 gene deletion may contribute to a person’s increased vulnerability to sudden cardiac arrhythmia-related death. It has been discovered that this deletion mutation causes autosomal dominant inheritance of cardiac arrhythmias in people with Smith-Magenis syndrome (i.e., specific mutations result abnormalities).

Sestrin-2 is a highly conserved stress-induced protein that is expressed in mammals. It is secreted by cells such as macrophages, T lymphocytes and epithelial cells. It has a paradoxical role in tumor suppression and progression. Its expression has regulatory properties and prognostic value in lung cancer cells. It has been identified as a potential therapeutic target for diseases associated with cell death, such as cancer and neurodegenerative disorders. By inhibiting caspase 3 activation, it can protect cells from programmed cell death and maintain healthy cellular function.

TYPICAL RESULTS

For this SESN2 ELISA kit it is recommended that a standard curve is generated for each assay carried out.
