LDHA ELISA Kit (L-Lactate Dehydrogenase A Chain)
INTENDED USE
Human LDHA ELISA kit can measure concentrations of L-lactate dehydrogenase A chain (LDHA, LDHM, cell proliferation-inducing gene 19 protein, LDH-A) present is plasma, serum, biological fluids or tissue homogenate samples.
INTRODUCTION
LDHA encodes the L-lactate dehydrogenase A chain, a homotetrameric enzyme that catalyzes the anaerobic conversion of pyruvate to L-lactate with concomitant nicotinamide adenine dinucleotide (NAD+) regeneration. This terminal glycolytic reaction constitutes the major pathway for metabolic flux during ischemia, sepsis, or other states of tissue oxygen debt. Moreover, aggressive tumor cells preferentially utilize cytoplasmic LDHA despite oxygen availability (“Warburg effect”), reflecting a proliferative switch toward lactate production over oxidative phosphorylation.
Ubiquitously expressed across tissues, LDHA can translocate to the nucleus and modulate transcription—potentially regulating glycolytic genes during early differentiation or in response to cellular stressors. Furthermore, via HIF1α activation, hypoxic conditions induce LDHA transcription to levels disproportionate with other glycolytic enzymes, indicating rate-limiting control over anaerobic metabolism. LDHA expression correlates with tumor progression, metastasis, and poor prognosis across diverse malignancies.
Selective inhibitors and monoclonal antibodies targeting LDHA elicit promising antiproliferative, proapoptotic, and antimetastatic responses in preclinical cancer models, likely by intervening in phosphoglycerate kinase-1 secretion or glutamine metabolism. Additionally, starving energy production via combined mitochondrial and LDHA blockade elicits profound oxidative stress. While pharmacological inhibition demonstrated safety in phase I trials, on-target muscle side effects remain a concern.

TYPICAL RESULTS
For this LDHA ELISA kit, it is recommended that a standard curve is generated for each assay carried out.
