INTENDED USE

Human semaphorin-3G ELISA kit can measure concentrations of SEMA3G (semaphorin-3G, sem2) present is plasma, serum, biological fluids or tissue homogenate samples.

INTRODUCTION

Discovered through its homology to viral semaphorins, the class 3 secreted glycoprotein semaphorin-3G (SEMA3G) critically regulates a multiplicity of physiological processes during embryonic development and in mature tissues. SEMA3G binds to neuropilin and plexin cell surface receptors, triggering activation of numerous downstream signaling effectors to exert effects on cell adhesion, cytoskeletal organization, integrin function, and cell survival, migration, and proliferation.

SEMA3G exhibits a dynamic spatiotemporal expression profile during embryogenesis. It functions through repulsive cues to guide the migration and connectivity of neurons and developing vasculature. SEMA3G-signaling confers segmental patterning information, with vital consequences for cardiac and skeletal morphogenesis. The multifaceted roles encompass regulation of lung branching, kidney organogenesis, and endocrine pancreas innervation.

Intriguingly, SEMA3G promotes attachment of select tumor cell populations while inhibiting migration and growth—possibly through MAPK/ERK or other pathways—in a cell type-specific manner. Loss of expression due to hypermethylation of its promoter associates with progression of diverse malignancies like breast, bladder, and nasopharyngeal cancers. Manipulating SEMA3G and its receptors might prove therapeutically efficacious to restrict metastatic tumor spread. Beyond cancer, altered activity likely contributes to other diseases involving impaired nervous or vascular system development, such as congenital cardiac defects. Elucidating the complex signaling networks downstream of semaphorin guidance cues could clarify molecular underpinnings for an array of abnormalities stemming from aberrant cellular navigation during embryogenesis or tissue repair processes.

For this SEMA3G ELISA kit, it is recommended that a standard curve is generated for each assay carried out.
