Chitinase-3-Like Protein 1 ELISA Kit (CHI3L1)

INTENDED USE

Human CHI3L1 ELISA kit can measure concentrations of CHI3L1 (Chitinase-3-Like Protein 1, GP-39, ASRT7, CGP-39, YYL-40, YKL-40, Glycoprotein 39) present is plasma, serum, biological fluids or tissue homogenate samples.

INTRODUCTION

CHI3L1, also known as chitinase 3 protein 1 or YKL 40 is a protein found in mammals that has been linked to processes such, as inflammation, cancer development and tissue changes. Interestingly even though it lacks chitinase enzyme activity it still plays a role in these processes. CHI3L1 is released by types of cells like macrophages and cancer cells and its levels have been observed to be higher in diseases associated with inflammation and alterations in the extracellular matrix.

Despite not having activity like chitinase proteins do CHI3L1 interacts with components of the matrix and sends signals inside cells. These signals may be involved in promoting cancer cell growth activating macrophages supporting blood vessel formation (angiogenesis) and influencing how tissues respond to mechanical strain and oxidative stress. However the specific molecular mechanisms through which CHI3L1 exerts its effects are still not fully understood.

In conditions characterized by inflammation and tissue changes such as arthritis, liver fibrosis, atherosclerosis, asthma, inflammatory bowel disease (IBD) and various types of cancer; elevated levels of CHI3L1 have been observed. Measuring circulating levels of CHI3L1 shows promise as a tool for predicting treatment responses and prognosis in cancers as well as chronic inflammatory conditions.

However there are still uncertainties regarding whether increased levels of CHI3L1 drive processes or if they represent a protective response or consequence downstream. In the context of cancer specifically higher levels of CHI3L1 generally correlate with prognoses; however knockout studies suggest that the protein may actually inhibit stages of tumor formation.

For this CHI3L1 ELISA kit, it is recommended that a standard curve is generated for each assay carried out.
