Asprosin ELISA Kit
INTENDED USE
Human asprosin ELISA kit can measure concentrations of asprosin present is plasma, serum, biological fluids or tissue homogenate samples.
INTRODUCTION
Asprosin is a recently discovered fasting-induced glucogenic protein hormone released by white adipose tissue. As a C-terminal cleavage product of profibrillin, it circulates in nanomolar concentrations in response to nutritional scarcity. By acting on hepatocytes to stimulate glucose release, it functions as a counterregulatory signal mobilizing hepatic glycogen stores to maintain serum glucose and insulin levels during fasting.
Mechanistically, asprosin binds to the G protein-coupled orexin receptor OX1R on hepatocytes, activating cAMP-dependent signaling cascades that increase glucose production through gluconeogenesis and glycogenolysis. This glucose then enters circulation, acting on pancreatic beta cells to maintain insulin secretion that regulates uptake by peripheral tissues. Thereby asprosin links adipose tissue nutrient sensing to glucose homeostasis during fasting by directing hepatic glucose output in accordance with supply and demand signals.
However, chronically elevated asprosin arises in contexts of insulin resistance such as diabetes and obesity. This excessive activation of glucose production contributes to fasting and postprandial hyperglycemia by overwhelming pancreatic capacity, representing a pathogenic mechanism underlying diabetic complications. Approaches targeting asprosin synthesis, release from adipose tissue, and receptor-mediated hepatic actions may help normalize glycemic control. Already, insulin sensitizers like rosiglitazone can attenuate circulating asprosin levels in rodent diabetes models. OX1R antagonists also show promise for selectively inhibiting asprosin activity without broadly compromising vital orexin signaling.

TYPICAL RESULTS
For this asprosin ELISA kit, it is recommended that a standard curve is generated for each assay carried out.
