INTENDED USE
Human sirtuin 1 ELISA kit can measure concentrations of SIRT1 (sirtuin 1, SIR2-like protein 1, homolg 1, EC 3.5.1, SIR2alpha) present is plasma, serum, biological fluids or tissue homogenate samples. This assay has a minimum sensitivity detection limit of 0.188 ng/ml.
INTRODUCTION
SIRT1, known as sirtuin 1, is a NAD-dependent deacetylase, it removes acetyl groups from target proteins to modulate their activation states in response to metabolic or environmental signals. The longevity and health-related effects of calorie restriction relate partly to SIRT1 activity enhancement.
SIRT1 deacetylates histones in chromosomal DNA as well as various transcription factors and co-regulators to influence gene expression patterns. This alters balances between anabolism and catabolism, oxidative stress responses, apoptosis signaling, and circadian behaviors in cells. For instance, SIRT-1 enhances fat mobilization, inhibits fat storage/growth pathways, stimulates mitochondrial biogenesis, reduces inflammation through transcription factor regulation, and beneficially impacts circadian clock maintenance – effects amplified by nutrient deprivation.
Such SIRT1 regulatory impacts likely contribute to improved health and survival observed with calorie restriction in some species. Boosting its activity pharmacologically aims to mimic such effects. Compound screening reveals small molecule sirtuin activating compounds (STACs) can enhance SIRT1 activity to realize therapeutic effects in metabolic, neurodegenerative, inflammatory, and aging-related disease models. However, designing isoform-selective SIRT-1 modulators has proved challenging. Off-target effects and incomplete understanding of downstream mechanisms still require further study.
