INTENDED USE
Human pleiotrophin ELISA kit can measure concentrations of PTN (pleiotrophin, OSF-1, HBNF-1, HB-GAM, HBBM, HBGF-8) present is plasma, serum, biological fluids or tissue homogenate samples. This assay has a minimum sensitivity detection limit of 18.75 ng/ml.
INTRODUCTION
PTN, or pleiotrophin, is a developmentally regulated protein that is highly concentrated in the brain and bone during neonatal development but expressed at lower levels in many tissues in adults. By binding cell surface receptors, pleiotrophin activates signaling influencing cell proliferation, migration, and survival.
Expressed early in development, it is critical for normal brain, bone, and epithelial cell growth. Through boosted protein and DNA synthesis, it enhances cell division rates and tissue expansion. It also participates in synapse formation and neural process growth. Following maturation, continued PTN expression facilitates wound healing responses in bone fractures and vascularization. However, overactive or prolonged PTN signaling is associated with pathological excess cell proliferation in some cancers and inflammatory disease progression by exacerbating abnormal angiogenesis and cell migration.
Pleiotrophin’s receptor binding activates major cellular pathways like MAPK and PI3K/AKT. This alters gene transcription to induce proteins enabling cell cycle progression and inhibiting apoptosis. PTN additionally interacts with extracellular matrix proteins and cytokines to stimulate changes benefitting cell migration, adhesion, and intercellular communication.
