INTENDED USE
Human ICTP ELISA kit can measure concentrations of cross-linked C-terminal telopeptide of collagen alpha-1 (ICTP) present is plasma, serum, biological fluids or tissue homogenate samples.
INTRODUCTION
ICTP, or cross-linked C-telopeptide of type I collagen, constitutes a peptide degradation product generated by osteoclastic resorption of mature, crosslinked collagen type I fibrils that make up the bone matrix. As bone tissue undergoes constant turnover mediated by opposing activity of osteoblasts and osteoclasts, biomarkers of the coordinated remodeling process provide critical insight into prevailing bone balance in research and clinical contexts. In particular, immunoassays detecting ICTP in serum or urine offer specific quantification of catabolic activity.
During the breakdown phase of bone remodeling handled by osteoclasts, proteolytic enzymes like cathepsin K hydrolyze the crosslinked collagen meshwork constituting the mineralized matrix. This liberates small peptide fragments from type I collagen in bone including ICTP, which then enter circulation for renal clearance. Since generation of this C-terminal telopeptide requires prior formation and maturation of crosslink-stabilized collagen before subsequent osteoclastic digestion, ICTP levels closely reflect bone resorption activity.
Correspondingly, elevated ICTP strongly associates with high-turnover osteoporosis, correlating with risk factors like postmenopausal status. ICTP also augments in conditions like hyperparathyroidism, Paget’s disease, and bone metastases exhibiting uncontrolled osteoclastic activity. As such, quantitative ICTP enzyme immunoassays facilitate disease screening, treatment monitoring to slow excess resorption, and fracture risk prognostication. Similar assays also show promise in assessing cartilage catabolism and rheumatoid arthritis disease activity representing other areas of collagen degradation.

TYPICAL RESULTS
For this ICTP ELISA kit, it is recommended that a standard curve is generated for each assay carried out.
