INTENDED USE
Human IL-33 ELISA kit can measure concentrations of interleukin-33 (IL-33) present is plasma, serum, biological fluids or tissue homogenate samples. This assay has a minimum detection limit of 9.375 pg/ml.
INTRODUCTION
Interleukin 33 (IL-33) is a molecule that acts as a warning signal, for tissue damage and alerts the system. It achieves this by binding to its receptor ST2 (IL1RL1) and the co receptor IL 1 receptor accessory protein (IL 1RAcP) which activates downstream signaling pathways like factor κB (NF κB). Mitogen activated protein kinase (MAPK).
When cells in our body are injured IL-33 is released passively from the nucleus of barrier cells such as epithelial cells. In cells it is anchored within chromatin by its C chromatin binding motif. However when cell death occurs due to necrosis it quickly diffuses out of the chromatin. Additionally under conditions bioactive full length IL-33 is actively secreted. Once released it affects types of cells including group 2 innate lymphoid cells T helper 2 lymphocytes, regulatory T cells, basophils, eosinophils, mast cells, macrophages, dendritic cells and natural killer cells. As a result of these interactions with cells IL-33 plays roles in type 2 inflammation that contributes to allergic diseases like asthma, atopic dermatitis and anaphylaxis.
However it’s important to note that IL-33 doesn’t solely promote inflammation. Its influence on lymphocytes helps suppress T cell responses and maintain tolerance towards harmless substances such, as food antigens or commensal microbes. Furthermore it prompts the release of cancer fighting cytokines, by killer cells and CD8+ T cells. As a result the signaling of IL-33 effectively maintains an equilibrium, between activation and regulation.
