INTENDED USE

Human TSG101 ELISA kit can measure concentrations of tumor susceptibility gene 101 protein (TSG101, ESCORT-I complex) present is plasma, serum, biological fluids or tissue homogenate samples.

INTRODUCTION

Tumor susceptibility gene 101 (TSG101) encodes a multifunctional protein best known for regulatory roles in cell growth, protein trafficking, and cancer progression when dysregulated. Identified originally as potential tumor suppressor in mammalian cells, subsequent insights revealed more complex functions dependent on cellular context. While frequently supporting cancer pathogenesis by enhancing proliferation and metastatic spread, TSG101 also serves vital roles in regulation of protein homeostasis through ubiquitin-mediated processing.

TSG101 participates in endosomal protein sorting as the central component of the ESCRT-I complex. By recognizing ubiquitinated membrane proteins marked for lysosomal degradation, ESCRT machinery clusters these cargo for involution into intraluminal vesicles budding into endosomes. TSG101 specifically binds ubiquitinated targets and recruits downstream ESCRTs enabling cargo capture and sorting into the MVB pathway terminating in lysosomal proteolysis. Thereby it critically maintains protein homeostasis by directing unwanted proteins towards degradation.

However in cancers, the same TSG101 activities enable growth-promoting effects and metastatic competence. By stimulating MVB formation, it enhances turnover of critical signaling proteins and receptors like epithelial growth factor receptor impacting downstream survival pathways. Furthermore, TSG101 overexpression in carcinomas promotes invasive behavior and intravasation. Hence, subversion of its protein trafficking functions in malignancy transforms this essential homeostatic regulator into a pathogenic enabler advancing disease severity.
