INTENDED USE
Human substance P ELISA kit can measure concentrations of SP (substance P) present is plasma, serum, biological fluids or tissue homogenate samples.
INTRODUCTION
Substance P is an undecapeptide that functions as a neurotransmitter and neuromodulator involved in transmitting signals for pain, stress, anxiety, nausea and inflammation. It belongs to the tachykinin family of peptides sharing a common C-terminal sequence. In the central and peripheral nervous system, substance P is produced from the preprotachykinin-A (PPT-A) gene and released from neuron endings to bind receptors including the neurokinin-1 receptor (NK1R).
As a key transmitter of nociception, substance P regulates pain signaling. Noxious stimuli trigger its release from primary sensory afferents, communicating with dorsal horn neurons of the spinal cord that modulate central pain pathways. This sensitizes second-order neurons, inducing hyperexcitability that amplifies pain. Its production and release further escalate in inflamed tissues. Consequently, substance P elicits neurogenic inflammation, directly stimulating immune cells and blood vessels to exacerbate swelling, vasodilation, and pain hypersensitivity. It likewise stimulates release of pro-inflammatory cytokines including histamine, serotonin, and prostaglandins.
Given its dominant roles in pain and neuroinflammation, substance P and its preferred NK1 receptor have emerged as promising therapeutic targets. NK1R antagonists help treat nausea, emesis, depression, and anxiety.
