INTENDED USE

Human IDO1 ELISA kit can measure concentrations of IDO1 (Indoleamine 2,3-Dioxygenase 1, IDO-1, 3-dioxygenase) present is plasma, serum, biological fluids or tissue homogenate samples. This assay has a minimum sensitivity detection limit of 0.094 ng/ml.

INTRODUCTION

Indoleamine 2,3 dioxygenase 1 (IDO1) is an enzyme, with effects. It plays a role in the step of tryptophan degradation along the kynurenine pathway. By reducing levels of tryptophan IDO1 acts as an immunosuppressant on T cells. Supports immune tolerance.

IDO1 can be found in tissues and white blood cells. It can be induced by signals related to inflammation such as interferons type I and II. During infection or inflammation IDO1 activity in antigen presenting cells like cells helps restrain T cell proliferation and activation in tissues. This effect occurs through the breakdown of tryptophan which’s an amino acid needed for T cell differentiation and expansion. The combination of tryptophan depletion and the production of apoptotic byproducts leads to a tolerant microenvironment that hampers T cell responses. Moreover high expression of IDO1 in antigen presenting cells is linked to increased populations of T cells that suppress inflammation.

In tumors IDO1 activity plays a role by promoting evasion and tolerance against anti tumor T cell responses. Many human cancers exhibit heightened levels of IDO1 expression, which is associated with infiltrating T cells and poorer prognosis. The catabolism of tryptophan mediated by IDO1 within tumor cells and tumor associated myeloid cells is crucial, for creating a tumor microenvironment that favors cancer progression. Inhibiting IDO1 could potentially reactivate tumor immunity.
