Flotillin-1 ELISA Kit (FLOT1)

INTENDED USE

Human flotillin-1 ELISA kit can measure concentrations of signal transducer and activator of FLOT1 (flotillin-1) present is plasma, serum, biological fluids or tissue homogenate samples.

INTRODUCTION

The FLOT1 gene encodes the 427 amino acid flotillin-1 protein, which is broadly expressed across human tissues but found at especially high levels in brain, heart, and hematopoietic cells. It contains a flotillin domain through which it can form oligomeric complexes with the related protein flotillin-2 within lipid rafts. Flotillin oligomerization helps stabilize lipid rafts and maintain their structural integrity. Flotillins also directly bind cholesterol and are implicated in cholesterol uptake and trafficking processes. Through these mechanisms, flotillins create specialized platforms that facilitate receptor recruitment into rafts and assembly of signaling molecules following receptor activation.

It is one of the major proteins found in lipid rafts – specialized areas within the cell membrane that contain high concentrations of cholesterol and glycolipids. flotillin-1 directly interacts with a variety of cell surface receptors and helps scaffold downstream signaling molecules.

Because flotillin is a key scaffolding protein within lipid rafts, it likely exerts functional effects on diverse cellular processes including cytoskeletal dynamics, membrane curvature and trafficking events, cell adhesion, and activation of proliferation pathways like MAPK signaling. While the precise regulatory roles of flotillin-1 remain to be fully characterized for many pathways, altered FLOT-1 expression has been reported in several cancers, and the protein may prove to have broad utility as a biomarker or therapeutic target.
