Collagen Type I Alpha 1 ELISA Kit (COL1A1)

INTENDED USE

Human COL1A1 ELISA kit can measure concentrations of COL1A1 (collagen type I alpha 1, COL1α1, alpha-1 type I collagen) present is plasma, serum, biological fluids or tissue homogenate samples.

INTRODUCTION

Collagen type I alpha 1 (COL1A1) encodes the pro-alpha1 chains of type I collagen, the predominant structural protein in the extracellular matrix of connective tissues including bone, tendon, ligament and skin. As such, it plays a pivotal role in tissues that endure mechanical stress such as bone, where type I collagen comprises 90% of the organic matrix. It forms the scaffolding that integrates the mineral phase conferring strength and resilience. Hence genetic defects in COL1A1 manifest in brittle bone disease such as osteogenesis imperfecta.

The COL1A1 gene resides on chromosome 17, containing a 50-kb coding region with 51 exons. It is extensively transcribed and translated, producing pro-alpha1(I) chains that enter the endoplasmic reticulum. Following folding and post-translational modification of lysine and proline residues, procollagen molecules comprised of two alpha1 chains and one alpha2 chain secreted into the extracellular space. There, cleavage of N- and C-terminal propeptides by specific proteinases yields insoluble collagen fibrils that hierarchically self-assemble into fibers and networks that offer tensile strength.

Beyond structural roles, type I collagen signaling via cell surface receptors including integrins, discoidin domain receptors and LAIRs regulates cell adhesion, proliferation, differentiation and survival. For example, changes in collagen density and crosslinking alter osteoblast signaling through β1 integrins and discoidin domain receptors. This evidence for “outside-in” signaling reveals extracellular matrix proteins like collagens are not passive scaffolds but directly communicate with residing cells to dynamicallyimpact tissue integrity and function.

For this COL1A1 ELISA kit, it is recommended that a standard curve is generated for each assay carried out.
