INTENDED USE

Human ALDOA ELISA kit can measure concentrations of aldolase A, fructose Bisphosphate (ALDOA) present is plasma, serum, biological fluids or tissue homogenate samples.

INTRODUCTION

As a tetrameric glycolytic enzyme catalyzing fructose-1,6-bisphosphate cleavage into dihydroxyacetone phosphate and glyceraldehyde 3-phosphate, fructose-bisphosphate aldolase A (ALDOA) holds a critical role regulating glucose metabolism and ATP generation through both catabolic and anabolic pathways. Mammalian aldolase exists as three isozyme subtypes with 65%-68% amino acid identity that differ in kinetic behavior, immunoreactivity, and predominant tissue expression patterns: ALDOA in muscle, ALDOB in liver, and ALDOC in brain.

While metabolically fundamental across cell types, ALDOA demonstrates the highest levels in adult striated muscle, consistent with the elevated glycolytic poise supporting contractile activity in these tissues. Beyond the cytosol, it also occupies non-canonical nuclear and sarcomeric locales. Through transcriptional cofactor functions or direct modulation of filament assembly dynamics, extracellular ALDOA further coordinates adaptive responses to hypoxic, oxidative, or mechanical stress. Dysregulation of ALDOA links with hereditary myopathies and glycogen storage disorders stemming from impaired muscle energy homeostasis.

Cancer cells share the upregulated glycolytic phenotype of contracting muscle, often with disproportionate elevation of ALDOA reflecting glycolytic dependence even under normoxic conditions. Thus, inhibiting elicits cytotoxic actions across numerous solid and hematological malignancies by triggering ATP crisis, oxidative damage, and apoptosis—particularly efficacious against metastasis. Impacts on cytoskeletal architecture may further mediate ALDOA blockade. Advancing knowledge of molecular switches dictating tissue-specific functionality should facilitate optimization of therapeutic targeting strategies.
