SARS-CoV-2 NP IgG ELISA Kit
INTENDED USE
Human SARS-CoV-2 NP IgG ELISA kit is intended for measuring IgG class of antibodies directed against nucleocapsid protein (NP) of the SARS-CoV-2 virus using samples of human serum, plasma and various other specimens.
INTROCUCTION
SARS-CoV-2 is short for severe acute respiratory syndrome coronavirus 2, it is virus that is composed from essentially four structural proteins. These proteins contain the membrane protein (M), ion channel envelope protein (E), the nucleocapsid protein (NP) and spike glycoprotein (S). The M protein is the first of the five proteins, it can form both surface and membrane-bound structures. The M protein connects to a portion of the RNA genome (the virus replicates by using viral RNA) and also attaches to other proteins that are present in the ribonucleoprotein complex. The second protein is E which stimulates cellular type 2A interferon signalling pathways, this helps regulate cytokines such as IL-2. It also binds to cell surface receptors that help cells recognize SARS-CoV. The third protein is NP which has two functions one being structural, while the other is to take up lipid. The fourth protein is the spike glycoprotein, this protein has a function of recognizing and binding to specific HLA antigens that are present on human cells.
The NP also binds double stranded ribonucleic acids (dsRNA) in order for them to be fully replicated and assembled into new virions. In doing this, it is able to bind two strands of dsRNA together in one conformation so that they are able to form helix and pore complexes through a 3D structure. The NP has also been found to have the ability of binding to membranes, promoting an endosome-to-virus trafficking pathway. The viral polymerase complex is believed to form the final structural protein in the virion, which is responsible for replicating new RNA molecules into DNA that will then be transcribed by the NP.

Blank Control: 0.117, 0.126 (OD450).
Positive Control: 0.737, 0.759 (OD450).
Serum From Healthy Subjects: 0.126, 0.15, 0.197 (OD450).
Serum From COVID-19 Patients: 1.373, 2.024, 0.582 (OD450).
Suggested Cut-Off Values:
Negative: < 0.20 (450nm).
Positive: ≥ 0.20 (450nm).
It is advised that each laboratory establishes its own normal and pathological refence range for detection of anti-NP IgG antibodies.
ASSAY CHARACTERISTICS
– Negative Result: Samples absorbance/cut-off < 0.9.
– Positive Result: Samples absorbance/cut-off > 1.1.
– Borderline Result: Samples absorbance/cut-off = 0.9 – 1.1.
– Sensitivity: 93.33%.
– Specificity: 93.89%.
