Sclerostin ELISA Kit (SOST)
INTENDED USE
Human sclerostin ELISA kit can be used for detecting concentrations of SOST (sclerostin) in human serum, plasma, biological fluids and tissue homogenates.
INTRODUCTION
Sclerostin (SOST) is a protein that is produced by osteocytes, these cells are mainly found in bone tissues. It is a family member of the inhibitory secreted frizzled-related proteins (SFRPs). It is a single polypeptide chain with a molecular weight of 25 kDa. SOST contains a cysteine-rich region that is predominately responsible for its inhibitory activity. towards the Wnt signalling pathway. Sclerostin is also produced in bone and other tissue cells by bone morphogenetic proteins (BMPs) including; BMP-2, -4, -5 and -7. This is a result of both genetic expression and mRNA translation.
One of the main functions of sclerostin is to act as an inhibitor to bone formation and it is able carry this out role by binding and inhibiting the activity of LRP5. Upon binding to LRP5, it is able to inhibit the activation of the Wnt signalling pathway, this will leads to affecting the downstream processes of bone formation and remodelling. The net affect will be a reduction in the formation of bone. Other important functions of SOST include regulation skeletal homeostasis and apoptosis. It is found to influence mineralization processes for example calcium deposition, phosphate deposition and soft tissue remodelling. The main function of sclerostin is the negative control of bone formation and remodelling process. It plays an important role in regulating the activation of Wnt signalling pathway.

The following SOST ELISA kit it is recommended that a standard curve is generated for each assay carried out.
